Nuôi cấy vi sinh vật để sản xuất các sản phẩm của các chất hoạt hoá sinh học là quá trình tinh vi và phức tạp nhất để thu nhận các sản phẩm tổng hợp vi sinh. Tổng hợp sinh học các chất hoạt hoá sinh học do vi sinh vật tạo ra phụ thuộc vào một số yếu tố như nhiệt độ, pH của môi trường và canh trường phát triển, nồng độ hoà tan, thời gian nuôi cấy, kết cấu và vật liệu thiết bị... Trong chương này chúng tôi sẽ giới thiệu một số thiết bị lên men công nghiệp được ứng dụng để cấy chìm vi sinh vật.

Nội dung:

Phụ thuộc vào các phương pháp ứng dụng để đánh giá hoạt động thiết bị lên men dùng để cấy chìm vi sinh vật và được chia ra một số nhóm theo các dấu hiệu sau:

Theo phương pháp nuôi cấy - các thiết bị hoạt động liên tục và gián đoạn.

Theo độ tiệt trùng - các thiết bị kín và các thiết bị không đòi hỏi độ kín nghiêm ngặt.

Theo kết cấu - các thiết bị lên men có bộ khuếch tán và tuabin, có máy thông gió dạng quay, có bộ đảo trộn cơ học, có vòng tuần hoàn bên ngoài; các thiết bị lên men dạng tháp, có hệ thông gió kiểu phun.

Theo phương pháp cung cấp năng lượng và tổ chức khuấy trộn, thông gió - các thiết bị cung cấp năng lượng cho pha khí, pha lỏng và pha tổng hợp.

Trong công nghiệp vi sinh thực tế hầu như tất cả các quá trình nuôi cấy sản xuất ra các chất hoạt hoá sinh học được tiến hành bằng phương pháp gián đoạn trong các điều kiện tiệt trùng.

Các thiết bị lên men nuôi cấy vi sinh vật trong điều kiện tiệt trùng

Nuôi cấy các vi sinh vật phần lớn được tiến hành trong các điều kiện tiệt trùng. Độ tiệt trùng của quá trình được đảm bảo bằng phương pháp tiệt trùng thiết bị lên men, các đường ống dẫn, cảm biến dụng cụ; nạp môi trường dinh dưỡng tiệt trùng và giống cấy thuần chuẩn vào thiết bị lên men đã được tiệt trùng; không khí tiệt trùng để thông gió canh trường và chất khử bọt tiệt trùng; các dụng cụ cảm biến tiệt trùng trong thiết bị lên men để kiểm tra và điều chỉnh các thông số của quá trình; bảo vệ vật đệm kín trục của bộ chuyển đảo, các đường ống công nghệ và phụ tùng trong quá trình nuôi cấy.

Thiết bị lên men có bộ đảo trộn cơ học dạng sủi bọt

Dạng thiết bị lên men này được sử dụng rộng rãi cho các quá trình tiệt trùng để nuôi cấy vi sinh vật - sản sinh ra các chất hoạt hoá sinh học.

Thiết bị lên men có thể tích 63 m3. Dạng thiết bị lên men này là một xilanh đứng được chế tạo bằng thép X18H10T hay kim loại kép có nắp và đáy hình nón (hình 10.1). Tỷ lệ chiều cao và đường kính bằng 2,6:1. Trên nắp có bộ dẫn động cho cơ cấu chuyển đảo và cho khử bọt bằng cơ học; ống nối để nạp môi trường dinh dưỡng, vật liệu cấy, chất khử bọt, nạp và thải không khí; các cửa quan sát; cửa để đưa vòi rửa; van bảo hiểm và các khớp nối để cắm các dụng cụ kiểm tra.
